After a single oral dose of trazodone-75 mg prolonged-release tablets of the Cmax of about 0.7 (g / ml are reached with a T max - 4 hours after administration, and an AUC of about 8 (g / ml / h. After a single oral dose of trazodone - 150 mg prolonged release, Cmax of about 1.2 (g / ml are reached with a T max - 4 hours after administration. The half-life of elimination is approximately 12 hours and AUC of about 18 (g / ml / h.

5.3 Preclinical safety data
Acute toxicity: LD50 of trazodone is orally administered 610 mg / kg in mice, 486 mg / kg in rats and 560 mg / kg in rabbits. The observed effects were: sleepiness, salivation, ptosis of the eyelids, and chronic convulsions. Subacute toxicity: subchronic studies were performed on rat, rabbit and dog, and chronic on rat, dog and monkey administered oral dose varies between 15 and 450 mg / kg / day in rats, 15 and 100 mg / kg / day in rabbits, 3 and 100 mg / kg / day in the dog and 20-80 mg / kg / day in the monkey. In studies in rats, the treatment caused hypertrophy of hepatocytes and soft endoplastichen reticulum followed by hepatomegaly. This effect is the result of detoxifying mechanism and could not be interpreted as pathological effect. Moreover lethal dose also produces effects already observed in acute toxicity tests. Dose / levels not observed adverse reactions / NOEL was 30 mg / kg / day. When rabbits were observed only effects of CNS depressants, with a corresponding NOEL 50 mg / kg / day. In the dog, the symptoms already seen in acute intoxication deteriorated in repeat dose and appropriate NOEL was 10 mg / kg / day. The monkey had more resistance from the dog and manifested only pharmacodynamic changes. NOEL was 30 mg / kg / day.
